1342 Spatial transcriptome profiling reveals TGFβ-2 in hair follicle progenitor cells is a potential driver of androgenetic alopecia

چکیده

Progenitor (PG) cells of the hair follicle are vital to growth and regeneration. Previous reports have highlighted their substantial reduction in from androgenetic alopecia (AGA) scalps. Since there is a limitation either investigating human PG native context or isolation forin vitro studies, no study focuses on molecular mechanisms underlying cell loss pathogenesis AGA . Here, state-of-the-art spatial whole transcriptome analysis regions revealed potential mechanism driving using intact scalp samples. NanoString GeoMxTM Whole Transcriptome Atlas (WTA) was used perform analysis. The gene expression profiles early (PG-A) control (PG-C) tissues were compared identify analyze major alterations, including biological processes candidate upstream regulators. Immunohistochemistry (IHC) with digital image validate results. A comparison between PG-A PG-C 197 differentially expressed genes. Pathway enrichment identified transforming factor beta (TGF-β) production as one significantly disturbed processes. QIAGEN Ingenuity Analysis (IPA) showed that TGFB2, an upregulated PG-A, regulator transcriptional alterations. Reassuringly, we detected increased TGFβ-2 immunostaining PG-A. Conclusively, profiling perturbation scalps microenvironment. We TGFβ-2might be player AGA.

Re-activation of Wnt/β-catenin Signaling Pathway in Hair Follicle Stem Cells in Treatment of Androgenetic Alopecia

Hair loss is a common hair disorder in human population. It affects quality of life and there are ongoing attempts to find permanent treatment for this condition. But, today there is no completely safe and protective treatment for all. Hair follicle stem cells are alive, but quiescence in androgenetic alopecia and are potentially active and can proliferate and differentiate, then regenerate hai...

Nestin expression in hair follicle sheath progenitor cells.

The intermediate filament protein, nestin, marks progenitor cells of the CNS. Such CNS stem cells are selectively labeled by placing GFP under the control of the nestin regulatory sequences. During early anagen or growth phase of the hair follicle, nestin-expressing cells, marked by GFP fluorescence in nestin-GFP transgenic mice, appear in the permanent upper hair follicle immediately below the...

Prostaglandin D2 inhibits hair growth and is elevated in bald scalp of men with androgenetic alopecia.

Testosterone is necessary for the development of male pattern baldness, known as androgenetic alopecia (AGA); yet, the mechanisms for decreased hair growth in this disorder are unclear. We show that prostaglandin D(2) synthase (PTGDS) is elevated at the mRNA and protein levels in bald scalp compared to haired scalp of men with AGA. The product of PTGDS enzyme activity, prostaglandin D(2) (PGD(2...

Inflammation in androgenetic alopecia

Years of research have led to the conclusion that androgenetic alopecia may be a result of an alteration in the hair growth cycle and/or a premature aging of the pilosebaceous unit. The etiology of androgenetic alopecia has been defined as multifactorial or even polygenic in nature. The fact that the success rate of treatment with either antihypertensive agents or modulators of androgen metabol...
